From a platform layout perspective, Everest Medicines is advancing the coordinated development of its proprietary AI neoantigen sequence prediction algorithm and mRNA/LNP technology, and has built a cutting-edge pipeline covering personalized neoantigen vaccines, universal tumor vaccines, immunomodulatory vaccines, and in vivo CAR-T.
EVEREST MED (01952) Hosts Another mRNA R&D Day as Its Proprietary Platform Enters the Clinical Validation Stage Recently, EVEREST MED (01952) held its 2026 mRNA Technology Platform R&D Day in Shanghai. This marks the second consecutive year the company has centered its R&D Day on its proprietary mRNA platform, disclosing technology iterations and pipeline progress in a concentrated manner. At the event, EVEREST MED systematically presented its mRNA/LNP technology platform, related R&D pipelines, and global R&D plans; Chairman of the Board Wu Yifang stated that the company aims to join the international first tier in the mRNA field. Shortly before the R&D Day, the world's first personalized neoantigen mRNA cancer vaccine, Intismeran Autogene, achieved a positive endpoint in Phase III clinical trials, enabling the entire field to cross a key validation threshold. For EVEREST MED's EVM16, its technological approach and clinical development strategy are similar to those of Intismeran Autogene; the latter's Phase III success as a global benchmark provides clinical validation and reference for this technological path. From a platform layout perspective, EVEREST MED is advancing collaborative R&D of its proprietary AI neoantigen sequence prediction algorithm and mRNA/LNP technology, and has established a cutting-edge pipeline covering personalized neoantigen vaccines, universal tumor vaccines, immunomodulatory vaccines, and in vivo CAR-T. Two former Moderna core scientists who were deeply involved in the R&D of Intismeran Autogene have joined the company, which is expected to further strengthen the company's global R&D capabilities and international collaboration layout for its mRNA technology platform. **Field Just Achieved Phase III Validation** Before the R&D Day, the clinical development of personalized neoantigen mRNA cancer vaccines had just crossed a key threshold. The INTerpath-001 Phase III study, presented at the 2026 European Society for Medical Oncology (ESMO) Annual Meeting, showed that Intismeran Autogene (V940/mRNA-4157) combined with pembrolizumab, used as adjuvant therapy for completely resected high-risk Stage IIbIV cutaneous melanoma patients, significantly improved recurrence-free survival (RFS) and distant metastasis-free survival (DMFS) compared with pembrolizumab monotherapy, reducing the risk of recurrence or death by 42% and the relative risk of distant metastasis or death by 38%. This is the first individualized neoantigen therapy to achieve a positive result in a Phase III trial. Industry observers note that these results, for the first time through a large-sample randomized controlled study, validated the synergistic potential of "personalized neoantigen vaccine + immune checkpoint inhibitor" in remodeling the tumor immune microenvironment and inducing long-term immune memory, bringing the clinical development of personalized neoantigen mRNA vaccines into a new stage. At the same time, the study population, tumor types, and treatment settings still have clear boundaries, and the clinical value of long-term clinical benefit still warrants continued attention and anticipation. In addition, Intismeran Autogene's exploration in adjuvant treatment after resection of pancreatic ductal adenocarcinoma is also advancing, providing new clinical research evidence for extending this technological path to other tumor types. Professor Shen Lin, Director of the Department of Gastrointestinal Oncology at Peking University Cancer Hospital and Chair of the Gastric Cancer Expert Committee of the Chinese Society of Clinical Oncology, and Professor Lu Shun of Shanghai Chest Hospital Affiliated with Shanghai Jiao Tong University School of Medicine and Director of the Shanghai Lung Tumor Clinical Medical Center, both believe that the development path of cancer therapeutic vaccines differs from traditional anti-tumor drugs, and that their mechanisms of action and immune response characteristics give early-stage and curative treatment settings exploration value. The two experts further noted that in addition to post-operative adjuvant therapy, expansion to earlier treatment settings such as neoadjuvant therapy, as well as exploring potential applications in first-line treatment, also warrants further evaluation through clinical research. For advanced patients, appropriate combination strategies and potential beneficiary populations need to be explored in light of tumor burden, immune microenvironment, and prior treatment history. For EVEREST MED's pipeline EVM16 in the same field, its technological approach and clinical development strategy are similar to those of Intismeran Autogene; the latter's Phase III success as a global benchmark provides clinical validation and reference for this technological path. As an mRNA personalized neoantigen cancer therapeutic vaccine independently developed by EVEREST MED, EVM16 has completed its first-in-human clinical data readout, showing good safety, tolerability, significant immunogenicity, and positive preliminary efficacy signals. Some data were presented at the 2026 American Association for Cancer Research (AACR) Annual Meeting. As the principal investigator of the investigator-initiated trial (IIT) for EVEREST MED's EVM16, Professor Shen Lin noted that the preliminary results of EVM16's first-in-human clinical trial in patients with advanced solid tumors showed good safety and tolerability, with preliminary immune activity signals observed, preliminarily validating the clinical value of EVM16 in activating tumor immunity through neoantigen-driven T-cell responses. "This gives us confidence in exploring this technological direction in gastrointestinal tumors. The key next step is to further accumulate clinical data and identify more suitable patient populations and combination treatment strategies." Professor Lu Shun stated at the R&D Day that mRNA personalized cancer therapeutic vaccines, as a novel treatment modality, have changed the approach to cancer treatment and provided a new technological path for tumor immunotherapy. He particularly emphasized that in early-stage cancer and curative treatment settings, for patients who have undergone surgery or other curative treatments but still face recurrence risk, how to translate immune responses into reduced recurrence risk is a direction that cancer therapeutic vaccines should focus on exploring. According to the company's disclosed R&D plans, EVM16 will add new indication expansion cohorts based on the Phase Ia study to further explore its clinical efficacy, and plans to submit a U.S. IND application in the first half of 2027. **"Joining the International First Tier in the mRNA Field"** From personalized neoantigen vaccines to universal tumor vaccines, mRNA technology is broadening the R&D path for tumor immunotherapy and gradually extending into areas such as in vivo cell therapy. At the R&D Day, Dr. Lakshmi Srinivasan, Senior Vice President and Head of Translational Medicine at EVEREST MED, and Dr. Brandon Coder, Vice President of Translational Science and Early Research, participated in forums and systematically presented on topics including the technological frontiers of mRNA cancer therapy, the construction of the proprietary AI + mRNA/LNP technology platform, R&D pipeline layout, clinical development of key projects, and manufacturing. Notably, both previously worked at Moderna and participated in the core R&D and clinical translation of the personalized neoantigen mRNA therapy Intismeran Autogene (V940/mRNA-4157). Their joining is also seen by observers as a signal of EVEREST MED accelerating its overseas clinical layout. Dr. Lakshmi Srinivasan, who previously served as Head of Oncology Translational Medicine at Moderna, noted that global mRNA cancer therapy R&D has formed multiple directions including personalized neoantigen vaccines, off-the-shelf tumor vaccines, and immunomodulatory vaccines; the field of in vivo mRNA applications is also moving from proof of concept to clinical practice. Combining global R&D progress, she specifically analyzed early exploration of related vaccines in solid tumors such as melanoma and pancreatic ductal adenocarcinoma, and noted that antigen selection, patient screening, delivery efficiency, and manufacturing are important links in driving clinical translation. Dr. Lakshmi Srinivasan stated, "mRNA/LNP technology is programmable and flexible in application, enabling exploration of multiple tumor treatment paths through different antigen designs and delivery strategies." In the future, the accuracy of antigen prediction, delivery efficiency, and the production cycle and delivery capability of personalized products will be key to driving clinical translation of the technology. EVEREST MED's R&D strategy aligns with global trends in mRNA/LNP therapies. From the existing layout, the company's proprietary mRNA platform supports multiple tumor vaccine products including personalized tumor vaccines, off-the-shelf TAA vaccines, and immunomodulatory vaccines (IM), with potential for combination with other treatment modalities as well as potential for combined use among themselves. The company's proprietary AI neoantigen sequence prediction algorithm can screen and predict potential neoantigens in patients' tumors. In addition to point mutations, insertions/deletions, and gene fusions, the algorithm is also exploring potential neoantigen sources such as alternative splicing, viral sequences, and endogenous retroviral sequences. EVEREST MED Chairman of the Board Wu Yifang stated, "mRNA is a frontier technology field with important strategic value. Since initiating our layout in 2021, the company has maintained strategic focus, continuously invested in R&D, and advanced platform construction. We have now formed an end-to-end technology platform covering AI algorithms, antigen design, delivery technology, process development, and manufacturing, which supports R&D in directions such as cancer therapeutic vaccines and in vivo CAR-T. In the future, the company will continue to invest in R&D, talent development, and technological innovation to drive iterative upgrades of its proprietary technology platform." He acknowledged that China has comprehensive advantages in clinical resources, investigator-initiated clinical research, professional talent, R&D efficiency, and manufacturing costs. Based on this, the company will integrate international scientific resources with China's clinical and industrial advantages in its globalization layout, promoting R&D achievements to international markets while consolidating its domestic leading position, and striving to join the international first tier in the mRNA field. Luo Yongqing, Chief Executive Officer of EVEREST MED, stated, "Over the past five years, the company's mRNA technology platform has gone through a development journey from technology introduction and accumulation of clinical development experience to independent R&D and continuous iteration. Through the global Phase II clinical study of the PTX-COVID19-B COVID-19 vaccine, the company accumulated experience in human immunogenicity, safety, and clinical development. Subsequently, the company further advanced independent technology R&D, continuously iterating algorithms and delivery systems, establishing end-to-end capabilities across the mRNA platform value chain, and achieving preliminary positive clinical progress in projects such as personalized cancer therapeutic vaccines. In the future, the company will continue to advance the construction of automated, modular production systems to shorten production cycles and optimize costs, providing support for global clinical development and subsequent industrialization of innovative therapies." Currently, EVM14 has received IND approval in both China and the United States, and EVM16 has entered early-stage clinical research. The company has also extended mRNA/LNP technology to the in vivo CAR-T field, delivering CAR mRNA through proprietary targeted lipid nanoparticles (tLNPs) to explore direct generation of CAR-T cells in patients, with related covering autoimmune diseases and oncology.
